The translocation creates a fusion gene composed of the 3’ end of ALK and the 5’ end of nucleophosmin gene. EML4/ALK fusion gene is observed in ca. 4% of non-small-cell lung cancer. Aberrant ALK activity is observed in other carcinomas as well. ALK is comprised by an extracellular domain, a single pass transmembrane domain and an intracellular kinase domain.
