EL222 has reversible, light-dependent DNA-binding activity. The protein bears a flavin mononucleotide (FMN chromphore). Upon illumination with blue light an adduct between <scene name='75/759048/Fmn_cys/1'>cysteine 78 and FMN</scene> is formed. This changes the protein conformation so that the previously sequestered DNA-binding domain is free to bind DNA.
