Vaccines against FMDV are used where the disease is endemic, however, vaccination is not permitted in many countries due to the potential risk for the escape of FMDV. Therefore, there has been interest in producing empty capsid particles for use of potential vaccines against FMD.

To solve this conundrum, different strategies must be analyzed to determine an optimal system for the production of FMDV empty capsid proteins with a limited amount of 3C protease that can stll process P1-2A, with out adverse expression effects. Low levels of 3C expression can be achieved by making mutations, which can then be tested through western blot to see if it is sufficient enough to process P1-2A.
