Function
[RT23_YEAST] Required for maintenance of mitochondrial DNA.[1] [RTC6_YEAST] Probable component of the mitochondrial large ribosomal subunit involved in ribosome biogenesis and translation. May be involved in a process influencing telomere capping.[2] [3] [4] [HIBCH_YEAST] Hydrolyzes 3-hydroxyisobutyryl-CoA (HIBYL-CoA), a saline catabolite. Has high activity toward isobutyryl-CoA. Could be an isobutyryl-CoA dehydrogenase that functions in valine catabolism. Also hydrolyzes 3-hydroxypropanoyl-CoA (By similarity). Has an indirect role in endocytic membrane trafficking. May have a function in protein biosynthesis in mitochondrial small ribosomal subunit.[5] [6] [RT17_YEAST] May have a meiosis-specific role as it accumulates during the middle stage of sporulation.[7] [RT24_YEAST] Involved in mitochondrial genome encoded proteins translation. [MRP10_YEAST] Involved in mitochondrial genome encoded proteins translation.[9] [COX24_YEAST] Involved in the splicing of the COX1 mRNA.[10] [FYV4_YEAST] Involved in telomere length regulation and required for survival upon exposure to K1 killer toxin.[11] [12] [MHR1_YEAST] Transcription factor involved in regulation of RNA polymerase II-dependent transcription. Also involved in regulation of mitochondrial DNA recombination, maintenance and repair, and generation of homoplasmic cells.[13] [14] [15] [16] [17] [18] [19] [RM36_YEAST] Involved in mitochondrial translation. [RMAR_YEAST] Essential for mitochondrial protein synthesis and required for the maturation of small ribosomal subunits.[23]

Mitochondria have specialized ribosomes (mitoribosomes) dedicated to the expression of the genetic information encoded by their genomes. Here, using electron cryomicroscopy, we have determined the structure of the 75-component yeast mitoribosome to an overall resolution of 3.3 angstroms. The mitoribosomal small subunit has been built de novo and includes 15S ribosomal RNA (rRNA) and 34 proteins, including 14 without homologs in the evolutionarily related bacterial ribosome. Yeast-specific rRNA and protein elements, including the acquisition of a putatively active enzyme, give the mitoribosome a distinct architecture compared to the mammalian mitoribosome. At an expanded messenger RNA channel exit, there is a binding platform for translational activators that regulate translation in yeast but not mammalian mitochondria. The structure provides insights into the evolution and species-specific specialization of mitochondrial translation.
