Assignment 8: IIIB: Central-loop: Map the interactions involving the central loop that stabilize quaternary structure. Explain how these interactions contribute to quaternary structure stability.[2][5]

1. Since an applet is not shown, please remember to use the prokaryotic GS not the human GS.
2. The first paragraph is to the point, but I cannot tell if you have too many or not enough WIKI applets yet. All points appear to be covered.
3. The information on the proteases shows scholarship, but I am not sure that the details are required. You might want to just state that the region is accessible to proteases as another indication that it is exposed. It is very interesting if the protease accessibility changes with covalent modification or with substrate binding.
4. You might be able to show solvent exposed regions of the synthetase in JMOL.
5. If this material is available, you might want to be a bit more specific on how the loop conformation changes when modified.
6. You can use PDBsum to show what interactions occur between the central loop and neighboring subunits to show it is an important quaternary interaction.

More Commets

Hi

1. Where is the applet?
2. First paragraph good 3. second paragraph needs work: do not make a good argument for using the passive site terminology. Why not stick to standard terms. GS is regulated by ADP-ribosylation .... when is GS modified in this manner--does it replace adenylylation?
4. second paragraph needs work--the protease susceptibility stills seems like a curiosity to me.
5. Third paragraph has good content.Hi

In general all and some extra stuff is here. I think the organization may need a bit more thought.

More Comments--you have time

Hi

1. The WIKIS need to be titled properly--not with TEXT to Be Displayed.
2. four different hydrogen bonding interactions TextToBeDisplayed of the quartenary structure of Glutamine synthetase.--this sounds awkward and I do not know what it means.
3. I am not sure The spatial orientation TextToBeDisplayed of the loops has been found to be less accessible for interactions to occur, contributing to the catalytic activity of the enzyme. means,?
4. Some of the WIKIS are outstanding.
